If you have trouble with the optical path, where the Objective appears off the optical axis and not centered in the cube herea are a few things you could check:
-
Make sure the Z-stage is correctly oriented and the right objective mount is selected.
-
Confirm that both the bottom and top puzzle pieces are properly placed and fixed. Proper alignment can only be achieved if everything is fully connected by the puzzle pieces.
-
Go through the entire beam path step by step—misalignment often starts earlier in the setup.
If you removed the FluoLED from the cube and reinserted it rotated by 90°, this might cause issues (we’ve seen this before). -
Check if the objective mount itself is misaligned by switching out the automatic z-stage with the manuel z-stage from the Core Box
-
If the objective mount itself is misaligned:
- Maybe the screws came loose—try tightening them.
- Try replacing the screws with longer ones and use washers to fine-tune the height (works if the mount seems too short)
- use the objective mount from the CoreBox
Hope it helps
